（四）透明
透明的目的是將脫水劑從材料中除去，使材料透明，增記折光系數(shù)；同時(shí)便于下步的浸蠟和包埋等程序（因?yàn)榫烤共荒苋芙馐灒?。透明劑是一種既能與脫水劑混合，又能與包埋劑混合的藥劑。常用的透明劑為二甲苯和綠方，適用原則也是逐級(jí)進(jìn)行，可減少材料收縮。一般步驟是：2/3純酒精+1/3二甲苯→1/2純酒精+1/2二甲苯→1/3純酒精+2/3二甲苯→二甲苯（兩次），每級(jí)停留時(shí)間0.5~2小時(shí)或更長些。二甲苯穿透能力強(qiáng)，但又宜使材料收縮變脆，使用時(shí)不能在其中停留時(shí)間過長。若材料放入二甲苯中時(shí)有白色混濁現(xiàn)象發(fā)生，說明材料脫水不*，應(yīng)把材料退回至純酒精中再行脫水。
（五）浸蠟
浸蠟的目的是逐漸除去透明劑,并為包埋劑所代替。常用的包埋劑是石蠟。浸蠟過程是使石蠟慢慢溶于浸有材料的透明劑中，溶解在透明劑中的石蠟漸漸深入到材料的細(xì)胞中去，zui后使透明劑*被石蠟取代，以便切片。
一般所用的石蠟分軟蠟（熔點(diǎn)40℃ 以下）和硬蠟（熔點(diǎn)40℃以上）。浸蠟過程一般是從低溫到高溫、從低熔點(diǎn)到高熔點(diǎn)逐漸進(jìn)行，這一過程不能操之過急，否則浸蠟不*影響切片。此外，軟材料可用熔點(diǎn)較低石蠟，較硬的材料可用熔點(diǎn)較高的石蠟；冬季可用熔點(diǎn)較低的石蠟，夏季可用熔點(diǎn)較高的石蠟。石蠟必須純凈，不含雜質(zhì)。整個(gè)浸蠟過程需在恒溫箱內(nèi)進(jìn)行（按需要調(diào)整溫度）。材料zui后一次二甲苯透明后即可浸蠟，浸蠟過程中逐漸使二甲苯揮發(fā)，并不斷加入純石蠟，直至石蠟飽和為止。浸蠟時(shí)間大約1~2天。

(D) transparent

The general procedure is: 2/3 pure alcohol +1/3 xylene → 1/2 pure alcohol +1/2 xylene → 1/3 pure alcohol +2/3 xylene → xylene (twice), each stage of stay Time 0.5 ~ 2 hours or longer. Xylene penetration ability, but should make the material shrinkage brittle, when used can not stay in it for too long. If the material into the xylene when white turbidity occurs, indicating that the material is not compley dehydrated, the material should be returned to pure alcohol dehydration again.
(E) Baptist wax
The purpose of the dip wax is to gradually remove the clarifier and replace it with the embedding agent. Common embedding agent is paraffin. Baptist waxing process is slowly dissolve the paraffin in the transparent material soaked in the paraffin dissolved in the transparent agent gradually into the material cells, and finally to make the transparent agent compley replaced by paraffin, in order to slice.
Paraffin wax generally used points (soft point below 40 ℃) and hard wax (melting point of 40 ℃ or more). Baptist waxing process is generally from low temperature to high temperature, from low melting point to high melting point gradually, this process can not be hasty, or not compley affect the slicing dip wax. In addition, the lower melting point of soft materials available paraffin, harder materials available higher melting paraffin; available winter melting lower paraffin, paraffin melting point in summer available higher. Paraffin wax must be pure, free of impurities. The whole process of waxing needs to be carried out in the incubator (adjust the temperature as needed). After the last time the material is transparent, xylene can be dipped in wax, the xylene is gradually volatilized during the dipped wax, and pure paraffin is continuously added until the wax is saturated. Baptist wax time of about 1 to 2 days.
